ABSTRACT

Introduction: The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has been posing a severe threat to global public health. Although broadly neutralizing antibodies have been used to prevent or treat corona virus disease 2019 (COVID-19), new emerging variants have been proven resistant to these antibodies. Methods: In this study, we isolated receptor binding domain (RBD)-specific memory B cells using single-cell sorting method from two COVID-19 convalescents and expressed the antibody to test their neutralizing activity against diverse SARS-CoV-2 variants. Then, we resolved antibody-RBD complex structures of potent RBD-specific neutralizing antibodies by X-ray diffraction method. Finally, we analyzed the whole antibody repertoires of the two donors and studied the evolutionary pathway of potent neutralizing antibodies. Results and discussion: We identified three potent RBD-specific neutralizing antibodies (1D7, 3G10 and 3C11) from two COVID-19 convalescents that neutralized authentic SARS-CoV-2 WH-1 and Delta variant, and one of them, 1D7, presented broadly neutralizing activity against WH-1, Beta, Gamma, Delta and Omicron authentic viruses. The resolved antibody-RBD complex structures of two antibodies, 3G10 and 3C11, indicate that both of them interact with the external subdomain of the RBD and that they belong to the RBD-1 and RBD-4 communities, respectively. From the antibody repertoire analysis, we found that the CDR3 frequencies of the light chain, which shared high degrees of amino acid identity with these three antibodies, were higher than those of the heavy chain. This research will contribute to the development of RBD-specific antibody-based drugs and immunogens against multiple variants.

ABSTRACT

Airborne diseases, including COVID-19, are transmitted by respiratory droplets, which makes the study of the evolution of these droplets important to control the transmission. However, the evolution of the droplets is complex, being a multiphase, polydisperse, multicomponent system undergoing evaporation. To numerically investigate such multiphase flows, there are mainly two approaches. One is the Eulerian–Lagrangian (E–L) approach, which is widely used due to its ability to trace the dispersion and evaporation of individual droplets. However, this approach generally has high costs and difficulty in post-processing. The other one is the Eulerian–Eulerian (E–E) approach, which, though having lower costs, is less adopted because of its failure to treat the features of polydispersity and evaporation. In order to take advantage of the low-costs of E–E approach, the population balance equation (PBE) is combined with the E–E approach to trace the polydisperse evaporating droplets. Two PBE solving methods, sectional method (SM) and quadrature based moment method (QBMM), are used and compared. The codes are developed based on the OpenFOAM library and their abilities to predict size changes of evaporating droplets, evolution of expelled airflow front, and aerosols concentration are assessed by using the experimental and numerical results in literature. Good agreements with the reported results are found, indicating the reliability of the CFD-PBE approaches. The SM and QBMM are finally applied in the transport of cough droplets in a 3D chamber. The suspending trends of small droplets and the falling trends of the large droplets are obtained by both methods. The droplets are found to be able to travel a distance longer than 2 m, which is valuable for the guidelines of social distancing. Additionally, the advantages and disadvantages of SM and QBMM are discussed.

ABSTRACT

The COVID-19 pandemic continues to threaten human health worldwide as new variants of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emerge. Currently, the predominant circulating strains around the world are Omicron variants, which can evade many therapeutic antibodies. Thus, the development of new broadly neutralizing antibodies remains an urgent need. In this work, we address this need by using the mRNA-lipid nanoparticle immunization method to generate a set of Omicron-targeting monoclonal antibodies. Five of our novel K-RBD-mAbs show strong binding and neutralizing activities toward all SARS-CoV-2 variants of concern (Alpha, Beta, Gamma, Delta and Omicron). Notably, the epitopes of these five K-RBD-mAbs are overlapping and localized around Y453 and F486 of the spike protein receptor binding domain (RBD). Chimeric derivatives of the five antibodies (K-RBD-chAbs) neutralize Omicron sublineages BA.1 and BA.2 with low IC50 values ranging from 5.7 to 12.9 ng/mL. Additionally, we performed antibody humanization on broadly neutralizing chimeric antibodies to create K-RBD-hAb-60 and -62, which still retain excellent neutralizing activity against Omicron. Our results collectively suggest that these five therapeutic antibodies may effectively combat current and emerging SARS-CoV-2 variants, including Omicron BA.1 and BA.2. Therefore, the antibodies can potentially be used as universal neutralizing antibodies against SARS-CoV-2.

ABSTRACT

BACKGROUND: The variants of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) harbor diverse spike (S) protein sequences, which can greatly influence the efficacies of therapeutics. Therefore, it would be of great value to develop neutralizing monoclonal antibodies (mAbs) that can broadly recognize multiple variants. METHODS: Using an mRNA-LNP immunization strategy, we generated several mAbs that specifically target the conserved S2 subunit of SARS-CoV-2 (B-S2-mAbs). These mAbs were assessed for their neutralizing activity with pseudotyped viruses and binding ability for SARS-CoV-2 variants. RESULTS: Among these mAbs, five exhibited strong neutralizing ability toward the Gamma variant and also recognized viral S proteins from the Wuhan, Alpha, Beta, Gamma, Delta and Omicron (BA.1, BA.2 and BA.5) variants. Furthermore, we demonstrated the broad reactivities of these B-S2-mAbs in several different applications, including immunosorbent, immunofluorescence and immunoblotting assays. In particular, B-S2-mAb-2 exhibited potent neutralization of Gamma variant (IC50 = 0.048 µg/ml) in a pseudovirus neutralization assay. The neutralizing epitope of B-S2-mAb-2 was identified by phage display as amino acid residues 1146-1152 (DSFKEEL) in the S2 subunit HR2 domain of SARS-CoV-2. CONCLUSION: Since there are not many mAbs that can bind the S2 subunit of SARS-CoV-2 variants, our set of B-S2-mAbs may provide important materials for basic research and potential clinical applications. Importantly, our study results demonstrate that the viral S2 subunit can be targeted for the production of cross-reactive antibodies, which may be used for coronavirus detection and neutralization.

ABSTRACT

BACKGROUND: Infectious diseases are a major threat to public health, causing serious medical consumption and casualties. Accurate prediction of infectious diseases incidence is of great significance for public health organizations to prevent the spread of diseases. However, only using historical incidence data for prediction can not get good results. This study analyzes the influence of meteorological factors on the incidence of hepatitis E, which are used to improve the accuracy of incidence prediction. METHODS: We extracted the monthly meteorological data, incidence and cases number of hepatitis E from January 2005 to December 2017 in Shandong province, China. We employ GRA method to analyze the correlation between the incidence and meteorological factors. With these meteorological factors, we achieve a variety of methods for incidence of hepatitis E by LSTM and attention-based LSTM. We selected data from July 2015 to December 2017 to validate the models, and the rest was taken as training set. Three metrics were applied to compare the performance of models, including root mean square error(RMSE), mean absolute percentage error(MAPE) and mean absolute error(MAE). RESULTS: Duration of sunshine and rainfall-related factors(total rainfall, maximum daily rainfall) are more relevant to the incidence of hepatitis E than other factors. Without meteorological factors, we obtained 20.74%, 19.50% for incidence in term of MAPE, by LSTM and A-LSTM, respectively. With meteorological factors, we obtained 14.74%, 12.91%, 13.21%, 16.83% for incidence, in term of MAPE, by LSTM-All, MA-LSTM-All, TA-LSTM-All, BiA-LSTM-All, respectively. The prediction accuracy increased by 7.83%. Without meteorological factors, we achieved 20.41%, 19.39% for cases in term of MAPE, by LSTM and A-LSTM, respectively. With meteorological factors, we achieved 14.20%, 12.49%, 12.72%, 15.73% for cases, in term of MAPE, by LSTM-All, MA-LSTM-All, TA-LSTM-All, BiA-LSTM-All, respectively. The prediction accuracy increased by 7.92%. More detailed results are shown in results section of this paper. CONCLUSIONS: The experiments show that attention-based LSTM is superior to other comparative models. Multivariate attention and temporal attention can greatly improve the prediction performance of the models. Among them, when all meteorological factors are used, multivariate attention performance is better. This study can provide reference for the prediction of other infectious diseases.

ABSTRACT

Porcine epidemic diarrhea virus (PEDV), an enteropathogenic coronavirus, has catastrophic impacts on the global pig industry. However, there are still no anti-PEDV drugs with accurate targets. G-quadruplexes (G4s) are non-canonical secondary structures formed within guanine-rich regions of DNA or RNA, and have attracted great attention as potential targets for antiviral strategy. In this study, we reported two putative G4-forming sequences (PQS) in S and Nsp5 genes of PEDV genome based on bioinformatic analysis, and identified that S-PQS and Nsp5-PQS were enabled to fold into G4 structure by using circular dichroism spectroscopy and fluorescence turn-on assay. Furthermore, we verified that both S-PQS and Nsp5-PQS PQS could form G4 structure in live cells by immunofluorescence microscopy. In addition, G4-specific compounds, such as TMPyP4 and PDS, could significantly inhibit transcription, translation and proliferation of PEDV in vitro. Importantly, these compounds exert antiviral activity at the post-entry step of PEDV infection cycle, by inhibiting viral genome replication and protein expression. Lastly, we demonstrated that TMPyP4 can inhibit reporter gene expression by targeting G4 structure in Nsp5. Taken together, these findings not only reinforce the presence of viral G-quadruplex sequences in PEDV genome but also provide new insights into developing novel antiviral drugs targeting PEDV RNA G-quadruplexes.

ABSTRACT

How much the vaccine contributes to the induction and development of neutralizing antibodies (NAbs) of breakthrough cases relative to those unvaccinated-infected cases is not fully understood. We conducted a prospective cohort study and collected serum samples from 576 individuals who were diagnosed with SARS-CoV-2 Delta strain infection, including 245 breakthrough cases and 331 unvaccinated-infected cases. NAbs were analyzed by live virus microneutralization test and transformation of NAb titer. NAbs titers against SARS-CoV-2 ancestral and Delta variant in breakthrough cases were 7.8-fold and 4.0-fold higher than in unvaccinated-infected cases, respectively. NAbs titers in breakthrough cases peaked at the second week after onset/infection. However, the NAbs titers in the unvaccinated-infected cases reached their highest levels during the third week. Compared to those with higher levels of NAbs, those with lower levels of NAbs had no difference in viral clearance duration time (P>0.05), did exhibit higher viral load at the beginning of infection/maximum viral load of infection. NAb levels were statistically higher in the moderate cases than in the mild cases (P<0.0001). Notably, in breakthrough cases, NAb levels were highest longer than 4 months after vaccination (Delta strain: 53118.2 U/mL), and lowest in breakthrough cases shorter than 1 month (Delta strain: 7551.2 U/mL). Cross-neutralization against the ancestral strain and the current circulating isolate (Omicron BA.5) was significantly lower than against the Delta variant in both breakthrough cases and unvaccinated-infected cases. Our study demonstrated that vaccination could induce immune responses more rapidly and greater which could be effective in controlling SARS-CoV-2.

ABSTRACT

BACKGROUND: The outbreak of the COVID-19 pandemic has caused extensive public health concerns, posing significant challenges to healthcare services. One particular area of concern is the mental health of patients with mental disorder, who are often a neglected group. The aim of this study was to investigate the prevalence of, and associated factors for symptoms of post-traumatic stress disorder (PTSD) among patients with mental disorder in China during the COVID-19 pandemic. METHODS: Self-reported questionnaires were distributed to patients in four psychiatric hospitals in Beijing, China, between April 28th and May 30th, 2020. Information regarding sociodemographic characteristics, COVID-19 related factors, support, psychosomatic factors, and PTSD symptoms were collected using a series of scales, such as the Impact of Event Scale-Revised, the 7-item Generalized Anxiety Disorder Scale, the 9-item Patient Health Questionnaire depression scale, and so on. Multivariate regression was used to identify factors related to PTSD symptoms. RESULTS: A total of 1,055 patients with mental disorder were included in the final sample. The prevalence of PTSD symptoms was 41.3%. Hierarchical linear regression demonstrated that fear of the pandemic and anxiety were shared associated factors for both symptoms of PTSD and their subscales. Additionally, age was an associated factor for the total PTSD (ß = 0.12, p < 0.01), intrusion (ß = 0.18, p < 0.001), and avoidance (ß = 0.1, p < 0.05) symptoms; depression was an associated factor for the total PTSD (ß = 0.13, p < 0.001), intrusion (ß = 0.11, p < 0.01), and hyperarousal (ß = 0.19, p < 0.001) symptoms. CONCLUSIONS: The prevalence of PTSD symptoms was high among patients with mental disorder during the COVID-19 pandemic in China. This study found that age, fear of the pandemic, anxiety and depression are significant associated factors of PTSD symptoms in patients with mental disorder during the pandemic. We call for higher awareness and introduction of PTSD interventions to relieve the psychological stress in these patients.

In order to explore the effects of COVID-19 control measures on the concentration and composition of PM2.5-bound polycyclic aromatic hydrocarbons (PAHs), and to better understand the sources of PM2.5-bound PAHs, PM2.5, samples were collected at two sites in urban and suburban areas of Shanghai before the lockdown, during the lockdown, after the lockdown in 2020, and during the same periods in 2019. The mass concentrations of 21 individual PAHs were determined via GC-MS analysis. While the COVID-19 control measures significantly reduced the absolute concentration of PM2.5-bound PAHs, they had no significant effect on their relative abundances, indicating that the significantly reduced traffic emission may not originally be the major source of PAHs in Shanghai. The differences in the composition of PM2.5-bound PAHs at three different lockdown-related periods may be caused by the gas-particle distribution of semi-volatile PAHs. The similarity in the composition of PM2.5-bound PAHs in different functional areas and different periods brings more uncertainties to the identification of PAH sources using the diagnostic ratios. During the lockdown period, the toxic equivalent concentration of PM2.5-bound PAHs in Shanghai was estimated to decrease by about 1/4, which still exhibits substantial carcinogenic risk upon exposure via inhalation.

BACKGROUND: A growing body of evidence indicates that the outbreak of COVID-19 has had a significant influence on individuals' cognition, emotion, and psychological health. This study aims to explore the effect of the association between time perspectives and self-control on the well-being and ill-being among college students in China during the COVID-19 pandemic. METHODS: We conducted an online survey involving 1,924 participants in mainland China during the outbreak of COVID-19. A series of self-rating questionnaires measuring the perceived impact of COVID-19, time perspectives, self-control, as well as the statuses of well-being and ill-being were administered. Multiple indirect effects of time perspectives and self-control on well-being and ill-being were analysed through structural equation modelling. RESULTS: The present-hedonistic time perspective (an orientation on immediate impulses of pleasure) meditated the effects of perceived impacts on both well-being and ill-being, and the future time perspective (considering the outcomes of actions and decisions) mediated the effects on well-being. Moreover, the mediating effects were further mediated by self-control. Specifically, the impact of the future time perspective on ill-being was fully mediated by self-control (ß = 0.01, p < 0.01). CONCLUSION: Based on the results, it is evident that the present-hedonistic time perspective, the future time perspective, and self-control are related to higher levels of well-being and lower levels of ill-being, thereby providing further insight into the theoretical framework of time perspectives during the COVID-19 pandemic. Additionally, our findings provide practical implications for psychological interventions during the on-going COVID-19 pandemic, focusing on the effects of time perspectives and self-control on the well-being and ill-being of different individuals.

ABSTRACT

The long-standing pathogen prevalence hypothesis suggests that collectivism can protect from epidemics and pandemics in terms of psychological well-being. However, studies exploring the protective mechanism induced when collectivism meets cultural tightness (the strength of social norms and tolerance for deviant behavior) are few. Therefore, this study aimed to investigate the protective effect of collectivism in detail considering loose and tight cultural contexts. The sample comprised 2001 Chinese participants (Mage = 18.41 ± 2.388 years;50.2% female). Moderated regression analyses indicated that more perceived risk of COVID-19 predicted severe mental health responses (i.e., depression and anxiety), collectivism moderated this positive relationship but individualism did not. Notably, the protective effect of collectivism is especially evident in tight cultures but ineffective in loose cultures. This study emphasized that the protective effects of collectivism on mental health during a pandemic should be considered within the framework of cultural tightness. This study’s findings may advance knowledge about the relationship between cultural type and mental health during epidemics.

ABSTRACT

The novel coronavirus disease (COVID-19) pandemic remains a global public health crisis, presenting a broad range of challenges. To help address some of the main problems, the scientific community has designed vaccines, diagnostic tools and therapeutics for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. The rapid pace of technology development, especially with regard to vaccines, represents a stunning and historic scientific achievement. Nevertheless, many challenges remain to be overcome, such as improving vaccine and drug treatment efficacies for emergent mutant strains of SARS-CoV-2. Outbreaks of more infectious variants continue to diminish the utility of available vaccines and drugs. Thus, the effectiveness of vaccines and drugs against the most current variants is a primary consideration in the continual analyses of clinical data that supports updated regulatory decisions. The first two vaccines granted Emergency Use Authorizations (EUAs), BNT162b2 and mRNA-1273, still show more than 60% protection efficacy against the most widespread current SARS-CoV-2 variant, Omicron. This variant carries more than 30 mutations in the spike protein, which has largely abrogated the neutralizing effects of therapeutic antibodies. Fortunately, some neutralizing antibodies and antiviral COVID-19 drugs treatments have shown continued clinical benefits. In this review, we provide a framework for understanding the ongoing development efforts for different types of vaccines and therapeutics, including small molecule and antibody drugs. The ripple effects of newly emergent variants, including updates to vaccines and drug repurposing efforts, are summarized. In addition, we summarize the clinical trials supporting the development and distribution of vaccines, small molecule drugs, and therapeutic antibodies with broad-spectrum activity against SARS-CoV-2 strains.

The Identification With All Humanity (IWAH) scale was designed to measure the extent to which an individual identifies oneself with all human beings. The current research aimed to conduct the validation of IWAH in a Chinese population and its convergent validity, as well as test the implications of IWAH in associations with help-seeking behaviour during COVID-19. A serial of three studies was conducted from September 1st 2020 to the end of October 2020. The series of studies included Study 1- Exploring the dimensions of the IWAH scale with a sample of 2,881 participants, Study 2- Confirmatory Factor Analysis for the Chinese IWAH dimensions with a separate sample of 6,667 participants, and Study 3- Role of the IWAH in the COVID-19 pandemic with a sample of 9,046 participants. Study 1 found the Chinese version of the IWAH scale to be a two-dimensional construct, with factor 1 - Bond with Humanity and factor 2 - Human Kinship. Study 2 confirmed the two-factor construct as found in Study 1. It also showed positive relations between IWAH and moral judgement, collectivism, nature connectedness, and negative relations with callousness, and having anxiety and depressive symptoms. Study 3 found that IWAH was negatively related to fear of COVID-19 and positively related to the likeliness of help-seeking. This is the first research to test the factorial structure of the IWAH scale in a Chinese population, with the adaptation showing good psychometric properties. The implication of IWAH on fear of COVID-19 and help-seeking provided further understanding of the possible practical value of IWAH during times of global stressful life events. Furthermore, study 3 is the first to explore how IWAH relates to anxiety, depression, and callousness.

OBJECTIVES: This study aimed to evaluate the effects of Chinese Medicine (CM) on the health condition of the post-COVID-19 patients, particularly with the CM Syndrome diagnosis and Body Constitutions (BC), as well as related clinical characteristics. METHODS: 150 participants who had COVID-19 and discharged from Hong Kong public hospitals were recruited. They were provided with three to six months of CM treatments, during which assessments were made per month and at follow-up on their CM syndromes, BC, lung functions, and other medical conditions. This study was divided into two parts: (1) Retrospective survey: medical history of participants during COVID-19 hospitalization was collected during the baseline visit; (2) Prospective observation and assessments: clinical symptoms, lung functions, and BC status were evaluated in participants receiving CM treatment based on syndrome differentiation and clinical symptoms. RESULTS: The median hospitalization period was 16 days. Symptoms were presented in 145 (96.6%) patients at the day they were diagnosed with COVID-19. Fever, fatigue, and dry cough were the most common symptoms, exhibiting in 59.3% (89 of 150), 55.3% (83 of 150), and 46% (70 of 150) participants, respectively. Among the 150 post-COVID patients, majority (71.3%) were of the two particular post-COVID CM Syndromes (Qi Deficiency of Lung and Spleen, and Qi and Yin Deficiency). Upon CM treatment, there was an observable increase in participants reaching a balanced BC (i.e. healthy body conditions). The increase was observed to be more prominent in those without the particular CM Syndromes compared to those with the CM Syndromes. Main clinical symptoms in participants with the CM Syndromes decreased upon CM treatment. Occurrence of fatigue also dropped after CM treatment though not all accompanied clinical symptoms were resolved fully. Further to the improvement in terms of CM assessments, lung functions of the participants were found to show improvement after treatment. Both the performance in 6MWT and scores in the LFQ improved upon CM treatments (P < 0.05). CONCLUSION: This study provided evidence for individualized CM treatment on COVID-19 rehabilitation concerning the clinical symptoms improvements, lung functions improvement, and achieving a balanced BC. It is believed that CM may be a key to further promote rehabilitation and resolution of residual symptoms. Long-term large scale follow-up studies on sub-categorising post-COVID patients according to different CM syndromes would be required to further elucidate treatment of persistent symptoms that may be associated with long-COVID.

ABSTRACT

An infodemic has accompanied the COVID-19 pandemic. This study explores whether overexposure to COVID-19-related information amplifies emotional distress. A cross-sectional online survey was conducted in China during the outbreak and after the peak of the pandemic (N = 1313). A latent moderated mediation model was built to analyze this relationship. COVID-19 information exposure was found to relate positively with emotional distress, and risk perception mediated the association between them. Additionally, psychological resilience moderated the mediating effects of risk perception. However, five factors of resilience differed in their moderating effects. This study offers theoretical and practical implications apropos clinical intervention and public health management.

ABSTRACT

During the COVID-19 pandemic, we are fighting not only the novel coronavirus, but also the "infodemic" induced by the pandemic. Therefore, it is urgent to explore approaches for enhancing individual immunity against science-related misinformation. We conducted a cross-sectional study to examine the relationship between public engagement with science (PES) and scientific information literacy (SIL) during the COVID-19 pandemic from college students (N = 8075) in China. The results showed that there was a significant difference between attitudes toward and activities of PES. More importantly, both PES activities and PES attitudes were found positively associated with SIL, especially the PES attitudes. The empirical study is significant in demonstrating the predictive effect of PES on individual ability to recognize science-related misinformation, which is crucial for mitigating harm from the "infodemic." Our study indicated that other than a science communication model in order to restore public trust in science, PES is promising to be incorporated into informal science education to facilitate individual SIL.

ABSTRACT

The unexpected outbreak of COVID-19 triggered fear and anxiety in the general population. Exercise was one of the most widely promoted methods to improve body function when socially restricted. This study aims to examine the role of exercise in relieving stressful mental health outcomes (anxiety and depressive symptoms) during the COVID-19 pandemic and explore the underlying mechanism from the perspective of hope, using a combination of goal-directed planning (pathways) and motivation (agency). A cross-sectional online survey recruiting 2390 Chinese participants was conducted during the COVID-19 pandemic in China. A series of questions and scales, including the self-designed exercise questionnaire, the Adult Dispositional Hope Scale, the Generalized Anxiety Disorder Scale-7 and the Patient Health Questionnaire-9, were used to measure exercise, hope, anxiety symptoms and depressive symptoms, respectively. A structural equation model was constructed to test the hypothesis that exercise benefits mental health outcomes through the mediating role of hope. Our results showed that exercise relieved stressful mental health outcomes via three paths: one direct path (ß = -0.077, 95% CI = (-0.138, -0.017), p < 0.01), one indirect path through hope of pathways thinking (ß = -0.046, 95% CI = (-0.064, -0.027), p < 0.001) and another indirect path through hope of agency thinking (ß = -0.060, 95% CI = (-0.081, -0.039), p < 0.001). Our results showed that exercise could alleviate stressful mental health outcomes by promoting both hope of pathway thinking and agency thinking. It provided practical insights into psychological prevention and intervention by means of exercise during the COVID-19 pandemic.

ABSTRACT

The COVID-19 pandemic continues to threaten human health worldwide, as new variants of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have emerged. Currently, the predominant circulating strains around the world are Omicron variants, which can evade many therapeutic antibodies. Thus, the development of new broadly neutralizing antibodies remains an urgent need. In this work, we address this need by using the mRNA-lipid nanoparticle immunization method to generate a set of Omicron-targeting monoclonal antibodies. Five of our novel K-RBD-mAbs show strong binding and neutralizing activities toward all SARS-CoV-2 variants of concern (Alpha, Beta, Gamma, Delta and Omicron). Notably, the epitopes of these five K-RBD-mAbs are overlapping and localized around K417 and F486 of the spike protein receptor binding domain (RBD). Chimeric derivatives of the five antibodies (K-RBD-chAbs) neutralize Omicron sublineages BA.1 and BA.2 with low IC50 values that range from 5.7 to 12.9 ng/mL. Additionally, we performed antibody humanization on a broadly neutralizing chimeric antibody to create K-RBD-hAb-62, which still retains excellent neutralizing activity against Omicron. Our results collectively suggest that these five therapeutic antibodies may effectively combat current and emerging SARS-CoV-2 variants, including Omicron BA.1 and BA.2. Therefore, the antibodies can potentially be used as universal neutralizing antibodies against SARS-CoV-2.
